In monkey models, incredibly inflammatory, no prolonged expression levels, hard to manufacture (expensive reagents, unreliable reactions like "witchcraft" in late 80s). Formulations worked in cell culture but not animals (aggressive white cell infiltrates in mice and monkeys). Abandoned for in vivo delivery due to unacceptable risk. VCL (company where original patents filed) also abandoned RNA, couldn't make it work. Discovered RNA/DNA alone more effective than positively charged lipids (now called lipid nanoplexes).
